Key Takeaways
- Parabilis Medicines shares rose 13% today after the company said it will present updated zolucatetide data in desmoid tumors at ESMO on Saturday, Oct. 24.
- Investors are buying ahead of a readout that includes the dose-optimization data meant to set the dose and schedule for a planned Phase 3 trial.
- The bar is high: as of February, 74% of desmoid patients with at least two follow-up scans had an objective response.
- Parabilis expects its $1.1 billion in cash to fund operations into 2030, so weak data would hurt hopes for the drug more than the company’s funding.
Parabilis Medicines (PBLS) shares are up 13% today. The company announced that the data behind its lead drug will get the big stage this weekend.
There are no new results yet. But the title of Saturday’s ESMO talk calls zolucatetide “a first-in-class direct B-catenin:TCF inhibitor.” A big part of the story is that this target went undrugged for decades.
What exactly happened
Before Monday’s open, Parabilis said Dr. Mrinal Gounder of Memorial Sloan Kettering will give an oral presentation of updated Phase 1/2 data in desmoid tumors. That’s a rare tumor that usually shows up in young adults, many of whom need treatment for years. The talk is Saturday at 8:45 a.m. ET.
The ESMO slot has been public since mid-September. The data cutoff is the new part. In August, Parabilis planned to show February’s data cut. Monday’s release puts the cutoff at June 22, so we get four more months of follow-up, plus the dose-optimization arm that will decide the dose and schedule for the Phase 3 trial.
The bar is already high
As of Feb. 16, all 25 response-evaluable desmoid patients had seen their tumors shrink. Among the 19 with at least two follow-up scans, 14 had an objective response, according to Parabilis’ IPO prospectus. RA Capital says nirogacestat, the only approved desmoid drug, posted a 41% response rate in its registrational trial.
That leaves two questions for Saturday: which dose goes into Phase 3, and whether those responses last.
Cash is the easy part
Parabilis can afford to find out. Cash and short-term investments jumped to $1.12 billion at the end of June after the company’s $770.5 million IPO…

…and management expects that to fund operations into 2030. Burn ran close to $50 million in the March quarter:

(The June quarter’s much smaller burn came in the same quarter Regeneron (REGN) paid Parabilis upfront for a research deal, so I wouldn’t treat it as the new pace.)
My view is that buying before the data makes sense here. The response rates are already strong, and with cash into 2030, even a messy dose readout won’t force Parabilis to raise money.
Of course, early responses in a small, open-label trial can fade, and a stock up 13% going into a Saturday readout has a lot priced in.
